Overview

UC4 CAR-T is an investigational allogeneic (universal), multi-antigen targeting chimeric antigen receptor T-cell therapy. It is engineered using "compound CAR" technology to simultaneously target four antigens highly expressed on acute myeloid leukemia (AML) blasts and leukemic stem cells (LSCs): C-type lectin-like molecule-1 (CLL-1), CD33, CD38, and CD123. By targeting multiple antigens, the therapy is designed to overcome the challenge of inter- and intra-patient tumor heterogeneity and prevent immune escape via antigen loss, a common cause of relapse in single-target CAR-T therapies. As a universal CAR-T product, it is derived from healthy donor T cells and modified (often via gene editing to knockout the T-cell receptor and/or HLA molecules) to prevent graft-versus-host disease (GvHD) and minimize host-versus-graft rejection, allowing for off-the-shelf administration.
