UCAR-T cells targeting B7-H3 is an allogeneic (universal) chimeric antigen receptor (CAR) T-cell therapy designed to target B7-H3 (CD276), a transmembrane protein overexpressed in various solid tumors, including high-grade gliomas and glioblastoma. Unlike traditional autologous CAR-T therapies, UCAR-T cells are derived from healthy donors and engineered to prevent graft-versus-host disease (GvHD) and host-mediated rejection, typically by knocking out the T-cell receptor (TCR) and HLA molecules. This specific program is being evaluated by the Second Affiliated Hospital of Soochow University in a Phase 0/1 trial for patients with relapsed or refractory advanced glioma, aiming to provide an off-the-shelf therapeutic option for this aggressive malignancy.
