Overview

UCART2 is an allogeneic, "off-the-shelf" universal CAR-T cell therapy engineered to target CD2-expressing malignant T cells. The therapy is designed by deleting both the CD2 antigen (to prevent fratricide, where CAR-T cells destroy each other) and the T-cell receptor (to prevent graft-versus-host disease). UCART2 has shown efficacy in preclinical models against T-cell acute lymphoblastic leukemia (T-ALL) and cutaneous T-cell lymphoma (CTCL), resulting in prolonged survival and reduced tumor burden in mice. Its activity can be enhanced by combination with rhIL-7-hyFc (a long-acting recombinant human interleukin-7), which increases CAR-T cell expansion and persistence. UCART2 represents a next-generation approach to the treatment of relapsed or refractory T-cell malignancies, offering scalability by enabling the use in multiple patients from a single donor source.[1][2][3][5]
