Overview

UCARTCS1A is an allogeneic, off-the-shelf chimeric antigen receptor T-cell (CAR-T) therapy developed to target CS1 (also known as SLAMF7), a surface antigen highly and consistently expressed on multiple myeloma cells. The product is manufactured from healthy donor T-cells that are gene-edited using TALEN technology to remove both the endogenous T-cell receptor and the CS1 antigen before introducing a CAR construct specific for CS1/SLAMF7. This design aims to prevent fratricide among CAR-T cells and reduce graft-versus-host disease risk. Upon administration, these engineered T-cells selectively bind to and lyse CS1-expressing tumor cells in patients with multiple myeloma. Clinical trials have shown early antitumor activity in heavily pretreated relapsed/refractory multiple myeloma patients[2][3][4][5]. The drug was developed by Cellectis.
