**UCARTFAP** is an investigational, preclinical allogeneic gene-edited CAR-T cell therapy from Cellectis designed to target fibroblast activation protein alpha expressed on cancer-associated fibroblasts within the tumor microenvironment. It is part of Cellectis' off-the-shelf UCART platform and has been described as a non-alloreactive, immune-evasive product candidate generated using TALEN gene editing, including disruption of the T-cell receptor to reduce graft-versus-host risk and beta-2 microglobulin knockout to reduce host immune rejection. The therapeutic concept is stromal depletion rather than direct tumor-cell targeting: by eliminating FAP-positive cancer-associated fibroblasts, UCARTFAP is intended to reduce desmoplasia, improve immune-cell infiltration, and sensitize stroma-rich solid tumors to subsequent tumor-antigen directed therapies such as mesothelin-targeted CAR-T cells. Publicly available information indicates development has remained at the preclinical stage, with reported in vivo work in stroma-rich solid tumor models including triple-negative breast cancer.
