**UCARTMESO** is an investigational allogeneic, off-the-shelf **CAR-T cell therapy** developed by **Cellectis** for **mesothelin-expressing solid tumors**. It is a **gene-edited anti-mesothelin CAR-T product** generated from donor T cells and engineered with a **second-generation mesothelin CAR** plus multiple knockouts including **TRAC**, **CD52**, and **TGFBR2**. The intended mechanism is selective recognition and killing of **mesothelin-expressing tumor cells**, while **TGFBR2 disruption** is designed to reduce inhibitory TGF-beta signaling within the tumor microenvironment and **CD52 disruption** supports use with CD52-directed lymphodepletion strategies. Publicly available data indicate potent preclinical antitumor activity in vitro and in vivo, including models of **pancreatic cancer** and **pleural mesothelioma**, and the program has been described as being in **preclinical development** with plans to advance toward first-in-human testing.
