UCB-NK cells are natural killer (NK) cells derived from human umbilical cord blood (UCB). These allogeneic cell therapies are developed for immunotherapy applications, particularly in oncology. Compared to peripheral blood NK cells, UCB-NK cells offer advantages such as easier collection, cryopreservation for off-the-shelf use, and a higher proportion of progenitor populations capable of differentiating into functional NK cells. However, they tend to be more immature and less cytotoxic than adult peripheral blood NKs unless activated or expanded ex vivo with cytokines like IL-2 or IL-15. Clinical trials have shown that expanded or genetically modified UCB-NK cell products can induce anti-tumor responses in hematologic malignancies and solid tumors with low risk of graft-versus-host disease. Mechanistically, these therapies act through direct cytotoxicity against tumor targets via secretion of perforins and granzymes as well as cytokine production (e.g., IFN-γ), and may be further enhanced by genetic engineering or combination with antibodies[1][3][6][7][9].
