UCB9608 is a potent, selective, and orally bioavailable inhibitor of phosphatidylinositol 4-kinase III beta (PI4KIIIβ). It exhibits an IC50 of 11 nM for PI4KIIIβ and inhibits the human mixed lymphocyte reaction (HuMLR assay) with an IC50 of 37 nM[3][7][8]. UCB9608 is selective for PI4KIIIβ over PI3KC2 α, β, and γ lipid kinases, showing minimal activity against other kinases in tested panels[3][7][8]. The compound demonstrates improved metabolic stability, excellent pharmacokinetic properties, and notable solubility. In vivo, UCB9608 can significantly prolong allogeneic organ engraftment and has been shown, in preclinical models, to facilitate donor-specific operational tolerance in organ transplantation while enabling antitumor immunotherapy[10]. UCB9608 was developed for potential use in immunosuppression/organ transplantation and may be investigated for solid organ transplant tolerance and immuno-oncology settings[10].
