UCCT-BCMA-1 is an investigational, cryopreserved autologous BCMA-targeted CAR T cell therapy engineered using a non-viral CRISPR-based approach to insert a 1XX-costimulated anti-BCMA chimeric antigen receptor into the TRAC locus of patient T cells, aiming to improve safety, potency, and persistence over conventional viral BCMA CAR constructs.[1][8][9] Developed at the University of California, San Francisco with CIRM support for relapsed and refractory multiple myeloma, the product consists of adult or tissue stem cell–derived autologous T cells that recognize and eliminate BCMA-positive myeloma cells after lymphodepleting chemotherapy.[1][8][9] The program is currently in late preclinical development with the goal of enabling an IND and a first-in-human phase 1 trial in patients with relapsed and refractory multiple myeloma who have limited access to existing BCMA CAR T cell therapies.[1]
