Ucenprubart (LY3454738) is a humanized immunoglobulin G4 (IgG4) monoclonal antibody that acts as an agonist of the human inhibitory checkpoint receptor CD200R. By binding to and activating CD200R, which is expressed on mast cells and basophils, ucenprubart induces a negative signal that functionally inhibits these immune cells. This mechanism targets the pathogenesis of chronic spontaneous urticaria and atopic dermatitis by reducing inflammation mediated by mast cell and basophil activation. Ucenprubart was developed by Eli Lilly for the treatment of moderate-to-severe atopic dermatitis and chronic spontaneous urticaria in patients inadequately controlled with standard therapies[5][7][10].
