UFOX is a combination chemotherapy regimen comprising UFT (Tegafur plus Uracil), Oxaliplatin, and Folinic Acid. Tegafur, a prodrug of 5-fluorouracil, acts as an antimetabolite by inhibiting thymidylate synthase and incorporating into DNA and RNA, thereby disrupting nucleic acid synthesis and function. Uracil enhances 5-fluorouracil's bioavailability by inhibiting dihydropyrimidine dehydrogenase, an enzyme responsible for 5-fluorouracil metabolism. Oxaliplatin, a platinum-based antineoplastic agent, induces DNA damage by forming DNA adducts, which leads to inhibition of DNA replication and transcription. Folinic acid (leucovorin) enhances the cytotoxic effects of 5-fluorouracil by stabilizing the binding of its active metabolite to thymidylate synthase. This regimen has been investigated as a first-line therapy for metastatic colorectal cancer.
