Drug intelligence / Profile preview
UFZ-HM is a biomimetic, pH-responsive nanoparticle system designed for the targeted treatment of glioblastoma (GBM). It consists of a zeolitic imidazolate framework-8 (ZIF-8) core loaded with ursolic acid (UA) and iron oxide (Fe₃O₄) nanoparticles, coated with a hybrid membrane derived from neutrophils and glioma cells. The hybrid membrane coating provides dual-targeting capabilities through inflammatory chemotaxis (via the neutrophil membrane) and homologous recognition (via the glioma membrane), enabling the nanoparticles to cross the blood-brain barrier (BBB) and target residual glioma cells. In the acidic tumor microenvironment, the ZIF-8 framework degrades to release its cargo. Ursolic acid induces tumor cell apoptosis, while Fe₃O₄ triggers ferroptosis via the Fenton reaction, resulting in synergistic anti-tumor efficacy.
