UI-201 (also known as CHP:NE1) is an investigational lymph node-targeting nanoparticulate peptide vaccine developed by United Immunity in collaboration with Kyoto University and Mie University. It is designed as a booster therapy to enhance the efficacy, safety, and in vivo persistence of T-cell receptor (TCR)-engineered T-cell therapies (such as TBI-1301) targeting the NY-ESO-1 tumor antigen. UI-201 utilizes United Immunity's proprietary T-ignite® (Myeloid Targeting Platform™) technology, consisting of a cholesteryl pullulan (CHP) nanogel (pullulan nanoparticle) that encapsulates a long peptide antigen derived from the NY-ESO-1 cancer/testis antigen, along with a CpG oligodeoxynucleotide (TLR9 agonist) adjuvant. Upon subcutaneous administration, the pullulan nanoparticles selectively target DC-SIGN-expressing dendritic cells and macrophages in the draining lymph nodes, promoting efficient antigen presentation and robust activation of NY-ESO-1-specific TCR-T cells. UI-201 is currently in Phase 2 clinical development for the treatment of synovial sarcoma and advanced soft tissue sarcomas.
