UK-396082 is a potent and selective small molecule inhibitor of activated thrombin activatable fibrinolysis inhibitor (TAFIa), developed by Pfizer as an oral antithrombotic agent[3][4][5][6]. By inhibiting TAFIa with high affinity (Ki ≈ 10 nM), it increases plasmin activity and promotes the breakdown of fibrin clots[6][8]. This mechanism was investigated for potential therapeutic use in thrombosis and pulmonary fibrosis. Preclinical studies demonstrated efficacy in animal models for reducing clot formation and renal fibrosis[8]. Clinical development reached Phase I trials but was discontinued without approval for any indication[3][5].
