Overview

Ulimorelin is a synthetic, small molecule ghrelin receptor agonist developed as a first-in-class treatment for gastrointestinal motility disorders such as postoperative ileus (POI) and diabetic gastroparesis. It acts by selectively activating the growth hormone secretagogue receptor type 1a (GHSR-1a), stimulating gastric motility and accelerating gastric emptying. Unlike some related agents, ulimorelin has little or no effect on growth hormone release in rats but does stimulate GH and IGF-1 in humans. Ulimorelin demonstrated potent prokinetic effects in animal models and early human studies, but failed to show efficacy in pivotal clinical trials for POI. The drug was primarily developed by Tranzyme Pharma and later Lyric Pharmaceuticals[1][2][3][5][7].
