ULK-101 is a potent and selective small molecule inhibitor of Unc-51 like autophagy activating kinase 1 (ULK1), the primary protein kinase that initiates the autophagy cascade. In oncology, ULK-101 is utilized to block cytoprotective autophagy, a survival mechanism that cancer cells exploit to resist metabolic stress and therapeutic interventions. Preclinical research, including studies presented at the AACR 2026 meeting, has demonstrated that ULK-101 is active against mesothelioma cell lines and acts synergistically when combined with CDK4/6 inhibitors such as abemaciclib. This synergy occurs because CDK4/6 inhibition often triggers compensatory autophagy as a survival response, which ULK-101 then effectively inhibits to promote cell death.
