Overview

Ulonivirine (MK‑8507) is a selective, potent, and novel non-nucleoside reverse transcriptase inhibitor (NNRTI) developed for the treatment of HIV‑1 infection. It acts by allosterically inhibiting HIV‑1 reverse transcriptase at the classic NNRTI binding site adjacent to the polymerase active site. Preclinical studies indicate that ulonivirine retains activity against common NNRTI resistance mutations such as Y181C and K103N and demonstrates strong antiviral effects. The drug was designed for once-weekly oral administration due to its favorable pharmacokinetics. Development was paused in November 2021 after reduced CD4+ T-cell and lymphocyte counts were observed in combination trials with islatravir[1][4][7].
