Drug intelligence / Profile preview
Ultra-rapid lispro (URLi) is an advanced formulation of insulin lispro designed to provide faster insulin absorption and improved postprandial glucose control. The formulation contains treprostinil, a prostacyclin analog that enhances absorption through local vasodilation, and citrate, which increases local vascular permeability to speed up insulin absorption[6][7]. This combination allows URLi to appear in the bloodstream approximately 1 minute after injection, with a significantly faster onset of action compared to conventional insulin lispro[7]. URLi maintains the same amino acid structure as insulin lispro, which has proline at position B28 and lysine at position B29 (reversed from human insulin)[7][9]. This structural modification reduces hexamer formation, allowing for more rapid dissolution and absorption. In clinical studies, URLi demonstrated superior control of post-meal blood glucose levels at 1 and 2 hours compared to Humalog, while providing similar overall A1C reductions[2][5]. Ultra-rapid lispro (URLi) is a novel insulin lispro formulation developed to more closely match physiological insulin secretion and improve postprandial glucose control compared to standard insulin lispro (Humalog). It contains additional ingredients that accelerate insulin absorption, resulting in faster onset of action and superior post-meal blood glucose reductions.
