Ultrafast insulin refers to a drug development program and specific product candidates (such as HTI-501) developed by Halozyme Therapeutics that combined recombinant human hyaluronidase (rHuPH20) with mealtime insulins like insulin lispro or insulin aspart. The rHuPH20 enzyme acts as a temporary, local spreading agent by degrading hyaluronan in the subcutaneous space, which increases the permeability of the tissue and allows the insulin to be absorbed into the bloodstream more rapidly. This ultrafast absorption profile is intended to more closely mimic the natural prandial insulin response, thereby improving post-meal glycemic control and reducing the risk of late postprandial hypoglycemia. Although Halozyme received government grants for this program, they ultimately discontinued internal development of their own insulin formulations to focus on licensing the rHuPH20 platform (ENHANZE technology) to other pharmaceutical companies.
