UM-101 is a small molecule inhibitor of the anti-apoptotic protein Mcl-1 (Induced myeloid leukemia cell differentiation protein Mcl-1), developed by researchers at the University of Michigan. It is designed to overcome resistance to BRAF and MEK inhibitors in metastatic melanoma by restoring the intrinsic apoptotic pathway, which is frequently dysregulated in cancer cells to promote survival. Preclinical studies have demonstrated that melanoma cell lines resistant to vemurafenib (a BRAF inhibitor) or pre-treated with MEK inhibitors show increased dependency on Mcl-1 for survival. UM-101 has shown significant synergy in inducing apoptosis when combined with these standard-of-care targeted therapies, suggesting its potential as a rational combination therapy to prevent or treat acquired resistance in melanoma.
