Overview

UM729 is a small molecule pyrimido-indole derivative used to enhance the ex vivo expansion and maintenance of human hematopoietic stem and progenitor cells (HSPCs). Identified through high-throughput screening, it works to prevent the spontaneous differentiation of both healthy hematopoietic stem cells (HSCs) and leukemia stem cells (LSCs). Mechanistically, UM729 modulates the Hippo signaling pathway by increasing YAP protein levels and acts synergistically with aryl hydrocarbon receptor (AhR) antagonists, such as StemRegenin 1 (SR1), to promote the self-renewal of CD34+ cells. It is a critical component in specialized culture media (e.g., modified-3a medium) used to prepare high-quality grafts for hematopoietic stem cell transplantation and gene therapy applications, including treatments for HIV and various hematological malignancies.
