Overview

Umbilical cord blood CD34+ cell-derived CD161- innate lymphoid cells type 1 is an experimental adoptive cell therapy being investigated for the treatment of acute myeloid leukemia (AML). This therapy involves the generation of a specific subset of type 1 innate lymphoid cells (ILC1s), characterized as Lin-CD127+CD161-CRTH2-CD117-, from healthy donor umbilical cord blood CD34+ hematopoietic stem cells. These ILC1s exert anti-leukemic effects by secreting Tumor Necrosis Factor alpha (TNFα) and Interferon gamma (IFNγ). TNFα inhibits the transition of leukemia stem cells (LSCs) into immunosuppressive macrophage-like cells, while IFNγ suppresses the differentiation of leukemic blasts. This approach aims to overcome the functional impairment of endogenous ILC1s observed in AML patients, thereby limiting leukemogenesis and potentially prolonging disease-free survival.
