Overview

Umbilical cord blood-derived NK cells are natural killer (NK) lymphocytes isolated and expanded from human umbilical cord blood. These allogeneic cell therapy products are developed as "off-the-shelf" immunotherapies for cancer and other diseases. Compared to peripheral blood or adult donor sources, umbilical cord blood provides a higher proportion of hematopoietic stem/progenitor cells and less mature immune effector populations with robust proliferative capacity and lower risk of graft-versus-host disease due to greater immune tolerance[2][5]. Cord-blood derived NK cell therapies can be used unmodified or genetically engineered (e.g., CAR-NK), sometimes expressing additional molecules such as IL-15 for enhanced persistence[3][7][8]. Their mechanism of action is primarily direct cytotoxicity against tumor or virally infected target cells via recognition of stress ligands, antibody-dependent cellular cytotoxicity (ADCC), and cytokine secretion. Clinical trials have evaluated these therapies in hematologic malignancies such as multiple myeloma[6], leukemia[3], solid tumors including non-small cell lung cancer[7], often in combination with other agents.
