Umbilical cord blood natural killer (NK) cells are an allogeneic cell therapy product consisting of NK cells isolated and expanded from umbilical cord blood. Unlike T cells, NK cells do not require HLA matching to prevent graft-versus-host disease (GvHD), making them a suitable off-the-shelf therapeutic option. These cells exert anti-tumor activity through the release of cytotoxic granules (perforin and granzymes) and the production of pro-inflammatory cytokines upon recognition of tumor-associated stress ligands or the absence of self-MHC class I molecules. Sun Yat-sen University Cancer Center is evaluating these cells in a Phase I clinical trial for children with high-risk, recurrent, or refractory neuroblastoma to assess safety, tolerability, and preliminary efficacy following standard multimodal treatments.
