Overview

Umbilical cord tissue-derived mesenchymal stem cells (UC-MSCs) are a population of multipotent neonatal stromal cells typically isolated from the Wharton's jelly or the perivascular region of the human umbilical cord. These cells are characterized by their ability to differentiate into various mesodermal lineages, including chondrocytes, osteoblasts, and adipocytes, and are highly valued for their potent immunomodulatory and anti-inflammatory properties. UC-MSCs exert their therapeutic effects primarily through the secretion of paracrine factors such as cytokines (IL-10, TGF-β), growth factors, and bioactive molecules (PGE2, IDO, HLA-G5), as well as through direct cell-to-cell contact with immune cells. They are capable of inhibiting T-cell and B-cell proliferation, suppressing natural killer (NK) cell activation, and inducing the formation of regulatory T-cells (Tregs). Due to their low immunogenicity and ease of isolation and expansion, UC-MSCs are being extensively investigated as an allogeneic cell therapy for autoimmune and inflammatory conditions, including rheumatoid arthritis, systemic lupus erythematosus, and osteoarthritis.
