Drug intelligence / Profile preview
UMG2-CD3 is a novel asymmetric (2+1) bispecific T-cell engager (BTCE) designed to target CD1a and CD3ε. Developed by researchers at Magna Græcia University with support from BiovelocITA, the antibody is constructed using knobs-into-holes technology. It incorporates a novel monoclonal antibody, UMG2, which binds to a previously uncharacterized epitope of CD1a, a glycoprotein highly expressed on cortical T-cell acute lymphoblastic leukemia (T-ALL) cells, healthy thymocytes, and Langerhans cells. By simultaneously binding CD1a on T-ALL cells and CD3ε on T-lymphocytes, UMG2-CD3 redirects T-cell-mediated cytotoxicity against leukemia cells, inducing T-cell activation, proliferation, and cytokine secretion. In preclinical studies, UMG2-CD3 demonstrated potent anti-tumor activity and significantly prolonged survival in humanized mouse models of T-ALL.
