Drug intelligence / Profile preview
umitrelimorgene autodencel (ITI-1000) is an autologous dendritic cell vaccine therapy designed to target the cytomegalovirus (CMV) pp65 viral antigen, which is highly expressed in glioblastoma multiforme (GBM) but absent in normal brain tissue. Developed using Immunomic Therapeutics' proprietary UNITE (UNiversal Intracellular Targeted Expression) platform, the vaccine consists of patient-derived dendritic cells electroporated with mRNA encoding the human lysosomal-associated membrane protein 1 (LAMP-1) fused to the CMV pp65 matrix protein (hLAMP:pp65). This fusion directs the antigen to the lysosomal compartment for enhanced MHC class II presentation, activating both CD4+ helper T cells and CD8+ cytotoxic T cells to mount a robust anti-tumor immune response. It is typically administered intradermally in combination with adjuvants such as GM-CSF (sargramostim) and tetanus-diphtheria (Td) toxoid preconditioning to enhance dendritic cell migration to draining lymph nodes.
