Overview

UNC4241 is a **pan-TAM small molecule inhibitor** that targets the TAM family of receptor tyrosine kinases: TYRO3, AXL, and MERTK. It is characterized by potent inhibition with IC50 values of 1.4 nM for MERTK, 5.4 nM for AXL, and 2.3 nM for TYRO3. UNC4241 has pharmacokinetic properties suitable for once-daily oral dosing in mice. It has demonstrated activity in tumor models by delaying tumor growth, promoting T cell infiltration, augmenting anti-PD-1 immunotherapy, and reversing the immunosuppressive functions of myeloid-derived suppressor cells (MDSCs). UNC4241 also inhibits FLT3 kinase. It is used in research settings to study the immuno-tolerant tumor microenvironment and cancer immunotherapy resistance[1][7].
