An unconjugated BCMA antibody is a monoclonal antibody designed to target the B-cell maturation antigen (BCMA; TNFRSF17), a cell surface receptor primarily expressed on malignant plasma cells in multiple myeloma. Unlike antibody-drug conjugates (ADCs) or bispecific antibodies, an unconjugated (or 'naked') antibody lacks a cytotoxic payload or a second binding domain. Its therapeutic potential typically relies on immune-mediated mechanisms such as antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), or the direct inhibition of BCMA signaling by blocking its natural ligands, BAFF and APRIL. In clinical and preclinical development, such as the studies involving Heidelberg Pharma's HDP-101, the unconjugated antibody is frequently used as a control to isolate the effects of the antibody's binding and immune recruitment from the activity of a conjugated toxin.
