Overview

The undecavalent pneumococcal-protein D conjugate vaccine is an investigational or theoretical pneumococcal conjugate vaccine designed to protect against 11 serotypes of Streptococcus pneumoniae. This type of vaccine uses purified capsular polysaccharides from 11 different serotypes, each chemically linked (conjugated) to a carrier protein—specifically, protein D, which is derived from non-typeable Haemophilus influenzae. The use of a carrier protein like protein D enhances the immune response compared to polysaccharide vaccines alone by inducing T-cell dependent immunity and promoting immunological memory. Such vaccines are intended primarily for the prevention of invasive diseases such as pneumonia, meningitis, and bacteremia caused by Streptococcus pneumoniae in children and adults. While several multivalent (including decavalent and dodecavalent) PCVs have been developed or are in development globally, the specific "undecavalent" (11-valent) formulation with protein D as a carrier has been studied but is not widely marketed or approved at this time[4][5][6][8]. GlaxoSmithKline (GSK) has previously developed Synflorix—a 10-valent PCV using protein D as one of its carriers.
