Overview

Undisclosed MASH bispecific siRNA refers to an investigational small interfering RNA (siRNA) therapeutic designed for the treatment of metabolic dysfunction-associated steatohepatitis (MASH), previously known as nonalcoholic steatohepatitis (NASH). The term "bispecific" suggests that this siRNA is engineered to target two distinct mRNA transcripts implicated in the pathogenesis or progression of MASH, potentially offering a more comprehensive gene-silencing approach than monospecific agents. While specific gene targets are not disclosed, recent research highlights genes such as PNPLA3 and HSD17B13 as key contributors to disease progression and common targets for oligonucleotide-based therapies in this indication[1][7][8]. The mechanism of action involves delivery of double-stranded siRNA into hepatocytes, where it is processed by Dicer and incorporated into the RNA-induced silencing complex (RISC). The guide strand directs RISC to complementary mRNAs, leading to their cleavage and degradation, thereby reducing expression of pathogenic proteins involved in liver inflammation and fibrosis[5][6][10]. This modality represents a precision medicine approach aiming at disease modification by directly downregulating drivers of hepatic injury.
