Overview

Unfractionated formalinized allergoid is a chemically modified allergen extract used in allergen immunotherapy (AIT) for the treatment of allergic diseases such as grass pollen allergy. Allergoids are produced by polymerizing native allergen extracts with aldehydes, most commonly formaldehyde. This process reduces the ability of the allergens to induce IgE-mediated hypersensitivity (allergenicity) while preserving or enhancing their capacity to induce immune tolerance and blocking antibody responses. The chemical modification involves covalent cross-linking between protein molecules, primarily through reaction with primary amines such as lysine residues, resulting in high-molecular-weight polymers that are less likely to trigger immediate allergic reactions but still stimulate regulatory T cell responses and production of protective antibodies like IgG4. Unfractionated refers to the use of these polymerized extracts without further size-based separation or purification steps[2][3][6][7][10].
