UniCAR-NK is an allogeneic **CAR-NK cell therapy** based on genetically engineered NK-92 cells that express the modular UniCAR receptor platform developed at Helmholtz-Zentrum Dresden-Rossendorf and advanced by GEMoaB. The receptor uses an extracellular single-chain variable fragment directed against the peptide epitope E5B9 together with CD28 transmembrane and costimulatory elements and a CD3 zeta signaling domain. UniCAR-NK cells do not directly bind tumor antigens; instead, they are activated and redirected by separate tumor-specific target modules that bridge the UniCAR receptor to cancer antigens such as GD2 or HER2. This modular on off switch design is intended to improve safety, controllability, and retargeting flexibility across solid tumors and hematologic malignancies, and the program has been described primarily in preclinical development.
