Drug intelligence / Profile preview
**UniCAR-T** is a modular, universal chimeric antigen receptor T cell (CAR-T) therapy platform designed to improve the control and safety of standard CAR-T approaches. Unlike conventional CAR-T cells that have fixed antigen specificity, UniCAR-T cells are engineered with an inactive CAR structure that can be reversibly switched “on” or “off” through administration of separate, target-specific adaptor molecules (target modules or TMs). These TMs cross-link UniCAR-T cells to tumor-associated antigens such as EGFR or CD98 and trigger immune effector functions only in their presence, providing a greater degree of specificity and allowing real-time control over T cell activity. This switchable design reduces the risk of long-term toxicities and severe side effects like cytokine release syndrome (CRS), since therapy can be rapidly terminated by withdrawal of the targeting module. The UniCAR system has demonstrated potent antitumor activity *in vitro* and *in vivo* against both radiosensitive and radioresistant cancer cells, including head and neck squamous cell carcinoma. Developers of the UniCAR system include research teams at Helmholtz-Zentrum Dresden-Rossendorf and Technische Universität Dresden[1][5].
