Universal anti-CD19 CAR-T cells, specifically referring to Gracell Biotechnologies' GC027 (TruUCAR-19), are an investigational, off-the-shelf (allogeneic) chimeric antigen receptor T-cell therapy designed to target CD19-expressing B-cell malignancies. Unlike traditional autologous CAR-T therapies that require manufacturing from a patient's own cells, GC027 is derived from healthy donor T cells. This approach utilizes the proprietary TruUCAR platform, which employs gene-editing technology to knock out the T-cell receptor (TCR) and HLA class I molecules to minimize the risk of graft-versus-host disease (GvHD) and host-versus-graft (HvG) rejection. This 'universal' design allows for immediate treatment availability and potentially more consistent product quality. GC027 is primarily being evaluated in clinical trials for patients with relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL) and B-cell lymphomas.
