Universal CD19 CAR-T cells refer to allogeneic, off-the-shelf chimeric antigen receptor (CAR) T-cell therapies designed to target CD19-expressing B-cell malignancies. Unlike autologous CAR-T therapies, which require harvesting and engineering a patient's own T cells, universal CAR-T cells are derived from healthy donors and genetically modified (often using gene-editing technologies like CRISPR/Cas9 to disrupt the T-cell receptor and HLA molecules) to prevent graft-versus-host disease (GvHD) and host-mediated rejection. This approach allows for rapid, scalable manufacturing and immediate availability for patients. In a clinical trial (ChiCTR1900027269) sponsored by the Medical Center of Hematology at Xinqiao Hospital, Army Medical University, universal CD19 CAR-T cells are being evaluated for their safety and preliminary efficacy in patients with relapsed or refractory B-cell leukemia or lymphoma.
