Overview

Unmodified haploidentical donor T cells are a cellular therapy product being investigated by the Dana-Farber Cancer Institute as a critical component of an experimental immunosuppression-free (IS-free) hematopoietic cell transplantation (HCT) protocol. This approach is designed for patients with relapsed/refractory or ultra-high-risk acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS). The therapy involves the infusion of these unmodified T cells, which contain T effector (Teff) cells, following a prior infusion of regulatory T-cell (Treg)-enriched donor cells. The unmodified T cells are intended to provide a potent graft-versus-leukemia (GVL) effect to eliminate residual cancer cells, while the engineered Treg-to-Teff ratio aims to prevent graft-versus-host disease (GVHD) without the need for standard post-transplant pharmacological immunosuppression.
