UNO-A001 is a bispecific T-cell engager (TCE) designed to target CD38-expressing hematologic malignancies. Developed by Unogen Biotech and Tavotek Biotherapeutics, the molecule features a novel structure consisting of a high-affinity humanized camelid VHH moiety targeting CD38 and a lower-affinity antibody Fab arm targeting CD3 on T cells. This differential affinity design is intended to maximize engagement with CD38-positive cancer cells while minimizing non-specific T-cell activation in the periphery. In preclinical studies, UNO-A001 demonstrated potent in vitro cytotoxicity against multiple myeloma cell lines and significant anti-tumor activity in xenograft models. It is currently a clinical candidate being prepared for testing in multiple myeloma, mantle cell lymphoma, T-cell lymphoma, and acute myelogenous leukemia.
