Overview

Unpulsed autologous dendritic cells are a cellular immunotherapy product consisting of mature dendritic cells derived from a patient's own peripheral blood leukocytes that have not been loaded (pulsed) with specific tumor antigens. In the clinical research context led by Dr. Annick Desjardins at Duke University, specifically the DERIVe trial (NCT03688178) for newly diagnosed glioblastoma, these cells are utilized as a control pre-conditioning agent. They are administered intradermally to the vaccine site prior to the administration of CMV pp65-targeted dendritic cell vaccines. The primary objective of using unpulsed DCs in this setting is to evaluate whether pre-conditioning the vaccine site with mature, non-antigen-loaded cells improves the migration and subsequent efficacy of the therapeutic antigen-loaded DC vaccines compared to other pre-conditioning methods like tetanus-diphtheria (Td) toxoid.
