Overview

Unpulsed dendritic cells (DCs) refer to dendritic cells that have not been exposed ("pulsed") to exogenous antigens or tumor lysates prior to administration. Dendritic cells are professional antigen-presenting immune cells capable of initiating and modulating adaptive immune responses by presenting antigens to T lymphocytes[4][5]. In their "unpulsed" or immature state, these DCs are highly efficient at capturing antigens but express low levels of co-stimulatory molecules and immunostimulatory cytokines[5]. Unpulsed DCs can induce immune tolerance by presenting self-antigens without activating signals, leading to T cell anergy or the development of regulatory T cells[5]. In experimental cancer immunotherapy settings, unpulsed DCs serve as a control group compared with antigen-pulsed (e.g., tumor lysate-loaded) DC vaccines; they may have limited direct anti-tumor activity but can still influence the immune environment[2][6].
