An unselected tumor cell vaccine is a type of cancer immunotherapy that utilizes the entire population of tumor cells from a specific cancer line or patient as the source of antigens, rather than isolating specific subpopulations such as cancer stem cells (CSCs). In the context of research conducted at the University of Michigan, it was used as a comparative control to evaluate the efficacy of cancer stem cell-enriched vaccines. The vaccine is typically prepared by inactivating tumor cells (e.g., through irradiation or lysis) and is often administered in conjunction with dendritic cells to stimulate a broad polyclonal immune response against various tumor-associated antigens. Preclinical studies in murine models of melanoma (B16-D5) and squamous cell carcinoma (SCC7) demonstrated that while unselected tumor cell vaccines can induce anti-tumor immunity, they are significantly less effective than vaccines specifically targeting the CSC population.
