**Upacaptamab uzedotin (MGC018)** is an antibody-drug conjugate (ADC) developed by MacroGenics targeting B7-H3 (CD276), a cell-surface protein overexpressed in various solid tumors including prostate, ovarian, lung, and head and neck cancers. The antibody binds to B7-H3, enabling internalization and release of its payload, a duocarmycin-based DNA-alkylating agent (DUA), which induces DNA damage, cell cycle arrest, and apoptosis selectively in tumor cells. It is designed for intravenous administration in advanced solid tumors, with ongoing clinical evaluation in phase 1/2 trials showing antitumor activity and a manageable safety profile.
