Overview

UPAO is a drug/device combination product developed by Parion Sciences for the treatment of upper airway diseases characterized by mucociliary dysfunction, such as Primary Ciliary Dyskinesia (PCD) and Chronic Rhinosinusitis. The product integrates a small molecule **epithelial sodium channel (ENaC) inhibitor** with Parion's proprietary **trans-nasal pulmonary aerosol delivery (tPAD)** technology. The tPAD system is designed to deliver aerosolized medication specifically to the nasal and sinus cavities during normal breathing, eliminating the need for patient coordination or breath-holding. The ENaC inhibitor component works by blocking the absorption of sodium and associated water from the airway surface, thereby increasing the hydration of the airway surface liquid (ASL). This restoration of hydration facilitates the clearance of mucus by the cilia, addressing the underlying pathophysiology of impaired mucus transport in the upper respiratory tract.
