UR-144 is a synthetic cannabinoid small molecule originally developed by Abbott Laboratories as a potential analgesic. It functions as a potent and selective full agonist of the peripheral cannabinoid receptor 2 (CB2), exhibiting a high affinity (Ki = 1.8 nM) compared to its significantly lower affinity for the central cannabinoid receptor 1 (CB1) (Ki = 150 nM). Although investigated for its potential therapeutic application in pain management due to the role of CB2 receptors in nociception, UR-144 never received clinical approval and was not formally marketed as a pharmaceutical. Instead, it became prevalent on the recreational drug market starting in 2012, often found as an ingredient in synthetic cannabis products (commonly labeled as 'herbal incense'). Due to its psychoactive properties and potential for abuse, UR-144 is currently a controlled substance in many jurisdictions, including the United States, where it was emergency scheduled by the DEA in 2013.
