Overview

Urolithin B is a natural intestinal microbial metabolite derived from the transformation of ellagitannins found in foods such as pomegranates and some nuts. It is a member of the coumarins chemical family and has the molecular formula C13H8O3. Urolithin B exhibits a range of biological effects including antioxidant, anti-inflammatory, pro-oxidant, antiestrogenic and estrogenic activities, depending on context. It has demonstrated pharmacological potential in preclinical models for diverse indications—such as reducing inflammation and cartilage degeneration in osteoarthritis, ameliorating cognitive deficits and brain aging, enhancing muscle mass and function, improving metabolic parameters (e.g., reducing blood lipids/glucose and adiposity), providing cardioprotection, modulating gut microbiota, protecting kidney function, and showing anticancer properties. Mechanistically, urolithin B acts by inhibiting the NF-κB signaling pathway (a central regulator of inflammation), increasing expression of sarco(endo)plasmic reticulum calcium ATPase 2 for cardiac function, and promoting SIRT1 activation, among other targets. Notably, it crosses the blood–brain barrier and may have neuroprotective effects. All current evidence of benefit is based on animal and cell-based studies; there is no established pharmaceutical product or clinical indication approved for human disease[1][2][4][6][7][10].
