Overview

Usistapide (also known as JNJ-16269110 and R256918) is a small molecule inhibitor of microsomal triglyceride transfer protein (MTTP). It was developed by Johnson & Johnson for the treatment of obesity and type 2 diabetes. Usistapide acts by inhibiting MTTP, an essential protein involved in the assembly and secretion of very low-density lipoproteins (VLDL) in the liver and chylomicrons in the intestine, thereby reducing lipid absorption and promoting weight loss. Clinical trials evaluated its safety and efficacy primarily in overweight or obese patients, with endpoints including changes in body weight, BMI, lipid levels, glucose metabolism parameters, and other metabolic markers. The drug was administered orally as capsules. Development has been discontinued[2][7][9].
