Overview

Usistapide (K-167) is a selective 5-HT4 receptor agonist developed by Kowa Company for the treatment of gastrointestinal motility disorders. By acting as a prokinetic agent, it stimulates 5-HT4 receptors on enteric neurons, leading to the release of acetylcholine and subsequent enhancement of the peristaltic reflex. This mechanism is intended to improve gastric emptying and intestinal transit in patients suffering from conditions such as functional dyspepsia and chronic constipation. Although it reached Phase 2 clinical trials in Japan, development appears to have been discontinued as it no longer appears in recent corporate pipelines.
