Overview

Usnoflast is a novel, orally available small molecule and selective NLRP3 inflammasome inhibitor discovered and developed by Zydus Lifesciences. Its mechanism of action is the inhibition of the NLRP3 inflammasome, a protein complex involved in triggering inflammation in response to danger signals. Overactivation of NLRP3 is implicated in several neurodegenerative and inflammatory disorders. Usnoflast has been investigated in various preclinical models of neuroinflammation, Parkinson’s disease, inflammatory bowel disease (IBD), and multiple sclerosis. Clinically, it is under development for amyotrophic lateral sclerosis (ALS), with orphan drug and fast track designations from the US FDA, and is in clinical development or studies for Cryopyrin-Associated Periodic Syndromes (CAPS), Parkinson’s disease, and ulcerative colitis. Usnoflast (ZYIL1) has demonstrated safety and tolerability in Phase I studies and is advancing through Phase II clinical trials in its target indications.
