USP11 siRNA is a small interfering RNA (siRNA) designed to target and knock down the expression of Ubiquitin Specific Peptidase 11 (USP11). USP11 is a deubiquitinating enzyme that has been implicated in various oncogenic pathways, particularly in stabilizing estrogen receptor alpha (ERα) and steroid receptor coactivator-3 (SRC-3/NCOA3) in estrogen receptor-positive (ER+) breast cancer. Knockdown of USP11 via siRNA has been shown to reduce cell proliferation, induce G2 cell cycle arrest and apoptosis, and abrogate ERα transcriptional activity in preclinical models. Delivery methods under investigation include encapsulation in the amphipathic cell-penetrating peptide RALA (developed in collaboration with pHion Therapeutics) and delivery via biocompatible freeze-dried collagen-based scaffolds.
