USP28 shRNA is a gene-silencing therapeutic candidate designed to target and reduce the expression of Ubiquitin Specific Peptidase 28 (USP28). USP28 is a deubiquitinating enzyme that plays a significant role in the progression of multiple myeloma (MM) by stabilizing KIF20B, which in turn activates the NF-κB signaling pathway. By utilizing short hairpin RNA (shRNA) to knockdown USP28, this approach induces cell cycle arrest at the G0/G1 phase and promotes apoptosis in MM cells. Preclinical research indicates that USP28 knockdown significantly reduces tumor burden in xenograft models and enhances the sensitivity of MM cells to the proteasome inhibitor bortezomib.
